I'm pretty confident the answer is no, but why not ask the experts opinion. I've always stayed on top of greasing gimbal bearing fusing the zerk valve on the transom bracket. I slip our boat in freshwater throughout season. I'm sure by trying to do this while submerged a small amount of water would enter valve. Any thoughts?

There is no need to risk that. The grease does not get used up anyway. In fact when you have been greasing yours for years like I have, there will be a pile of grease in there to clean out when you replace the bellows.
Now on the other hand, the grease on the driveshaft splines does not last all that long. This is why I have advocated the yearly drive pull, inspection and lubrication. The best thing you can do for your sterndrive is regular service, checking for water in the bellows, smooth operation of the gimble bearing and u joints. This, and checking for water in the gear oil will add years and years to the life of the sterndrive.

But if you really think it needed it, I think you could do it without getting water in. If you had a mound of grease protruding a little in front of your grease gun head before you snapped it on, then I don't see how any water could get in (as long as the main body of your grease gun stayed above water)
