What year?
One should be the holding tank pump out (it should be labeled waste). Should be up on the top side rail.
One should be fuel (it should be labeled gas). Should be down on the side.
On the port side should be the water fill (should be labeled water). Should be up on the top side rail.

It will depend on the year, the 1999 user's manual shows all the fittings on the starboard side for the 278. Also you'll have to determine if your boat has a grey water system. Congrats on the boat and welcome to the site, there are a lot of very knowledgeable people here. As you're trying to determine what's what on the boat, you can go on to the four winns website and download the user manual for your specific year 278. People on here are happy to help, but you might get answers quicker than waiting for a response on here.

The 2 fillers are on the top and marked "waste" so obviously the pump out point and the other is "water". The fuel fill is lower on the outside and there is another "water" on the port side. It is a 2008 Vista 278. Great boat so far.

Ok so one of the "waste" ones will be the pump out from the head, and the other will be the pump out for the grey water system (I think, I've never had a boat with a grey water system). The one labeled water is the one you will want to fill for your fresh water holding tank on the boat.

On our 248, we had (1) waste and (2) water caps. The waste was obvious but the (2) water caps filled the 2 water tanks I had. 1 tank was for fresh water (galley, shower, etc.) and the second was used to flush the head (using fresh water vs. raw water).

Once again you don't say what year but on my 2008 278 the front starboard is gray water which I no longer use. The rear is the black water waste tank from your head. The fuel is on the starboard side clearly marked and quite prone to back flow when it gets full. The fresh water is on the port side by itself and mine is very near my sink located there.
